A microplate version of the traditional "pour plate" Ames Test.  The Muta-ChromoPlateTM Kit is sensitive and user friendly. It is commonly used to detect mutagenic potential of individual chemicals or mixtures of chemicals in water, wastewater, sediment, air aqueous extracts, new chemicals, foods, cosmetics, pharmaceuticals and any other material that can be put into aqueous solution or micro-suspension, such that it can be taken up by the test strain.

The Ames ISO Basic Kit is a fluctuation assay verison of the well-known “Ames Test” which uses a 48-well sample exposure format on a 384-well microplate to reduce reagent and plastic used.  The Ames ISO procedure was developed to test mutagenic materials in a variety of sources like environmental air and water samples, foods, personal care products, and industrial effluents by the fluctuation assay which is a variation of the traditional “Ames Test”.

The Ames ISO method provides researchers with a reliable test method which is facile to set up and run, amenable to samples in aqueous solution or suspension and produces an easily-detectable colorimetric endpoint after 2-3 days of incubation at 37 °C.

This Ames test method is a modification of the Ames ISO fluctuation assay. Bacterial strains are pre-exposed to differing concentrations of the mutagenic compounds with a growth media in suspension for 100 minutes. Each dilution is run in triplicate.

The samples are then transferred, mixed with a reversion solution and plated onto a 96-well plate while using a 48-well evaluation format. The samples are incubated for 2-3 days and the number of revertant colonies (individual wells) are counted by colorimetric analysis

A microplate toxicity bioassay, EBPI has developed the UMU-CHROMOTESTTM into a simple procedure based upon the International Organization for Standardization protocol ISO 13829 (Water Quality- Determination of the genotoxicity of water and waste water using the umu-test), which can be performed easily in a non‑specialized laboratory.

The UMU-ChromoTest™ (umuC assay) employs Salmonella typhimurium TA 1535 [pSK 1002] bacteria in which the SOS DNA repair response umuC gene has been linked to the β gal gene responsible for producing the β-galactosidase enzyme. The degree of DNA damage repair using the SOS gene pathway is directly linked to the production of β-galactosidase, which is measured by the enzyme’s reaction with a yellow chromogen. Strain TA 1535 [pSK 1002] contains both the rfa mutation, which produces a defective, more permeable cell membrane, and the uvrB mutation, which eliminates the accurate excision DNA repair mechanism, resulting in more repair by error-prone mechanisms. The results from this assay agree very closely with traditional Ames mutagenicity tests, with the added advantage of only using a single bacterial strain.
The assay is based on the Organization of Standardization Protocol ISO 13829.

7031 SOS Direct Contact Sediment Genotoxicity Test

The SOS line of assays are designed for rapid detection of genotoxicity or DNA damage and utilizes the synthesis of the B-galactosidase enzyme whose gene (LacZ) is connected to an SOS promoter. Once a lesion has been detected, the SOS promoter is induced to start the transcription of the SOS genes and the LacZ gene is transcribed as well. This gene produces an enzyme which is assayed using a simple colour change. The degree of colorimetric expression is quantified and given as the SOS-inducing potency (SOSIP). Although the SOS tests also correlate well with the Ames assays, they have been shown to be slightly less sensitive, although they also report less false-positive results.

A microplate toxicity bioassay to determine acute and chronic toxicity in water, effluents, and other liquids.  The Toxi-ChromoTestTM procedure exposes the bacteria to the toxicants in the sample for a short (90-minute) incubation period. After the incubation period, a chromogenic substrate is added. If the sample is toxic, no colour will develop and if the sample is non-toxic, a distinctive blue colour quickly develops. The kit provides a clear, completely objective measurement of the toxicity of the sample by a simple visual qualitative evaluation of the colour obtained, or quantitatively by spectrophotometry using a micro-plate reader.

The assay allows the bacteria to grow in direct contact with the toxicants in the sample, eliminating expensive extraction procedures. The endpoint of the assay is an easy to interpret colour reaction. If the sample is toxic, no colour will develop; if the sample is non-toxic, a distinctive blue colour develops around the sample.

The SediTox™ kit is sensitive to a wide spectrum of toxic substances such as heavy metals, organic and inorganic pollutants, antibiotics and priority contaminants. The assay detects toxicity directly without labour intensive solvent extraction procedures. The sensitivity of the assay combined with its ease of use make it an essential research tool that can be used qualitatively for field measurements and quantitatively in the laboratory to compare relative toxicity between samples. The assay is based on the ability of a toxicant to inhibit the de novo synthesis of an inducible enzyme (β-galactosidase) in a highly permeable mutant of E. coli and the results manifest as an easily observable colour change after a short incubation period.
